By using \color<3-4>{blue}{text}, the text will be blue on slides 3 and 4, and black on other slides. Is it possible to make the text red on slide 1 at the same time?

\color<beamer:2|handout:0>{red}{some text}
The thing to remember about overlay specifications is that the default assumption is to take the action. So when in handout mode, TeX sees \color<beamer:2>{red}{some text} and asks "I'm in handout mode, is there any reason not to execute \color?". To which the response is "No" as there's no specific handout declaration. Putting in handout:0 says "In handout mode only do this on slide 0". As there is never a slide 0, this effectively says "In handout mode don't do this".

When you call a \color<overlay>[colorsys]{color} command that changes the color from then on (within the current group) as it takes as argument only the color specification and not the content that needs to be colored.
What happens here is that the sub-itemizes are in their own color environment so they are not affected by your command; instead the other items are in the same color environment.
A simple example to illustrate the issue:

You can also use the \textcolor<overlay>[colorsys]{color}{content} macro which applies the change in color only to the content argument.
